The Italians overwhelmed Hungarian Timea Babos and Frenchwoman Kristina Mladenovic under the roof on Centre Court, claiming a 6-1 6-3 victory.
Vinci fired down a service winner to clinch it, prompting Errani to fall to the turf in celebration. Vinci embraced her before the pair raced up to the players' box to embrace friends and family.
Errani and Vinci have been the dominant doubles team in the women's game since 2012, the year in which they won the French Open and US Open. They added the Australian Open title last year and made a successful defence in January.
Saturday's Wimbledon victory makes them the fifth women's team to complete the career Grand Slam, and the first since Serena and Venus Williams achieved the feat in 2001.
Martina Navratilova and Pam Shriver remain the only combination to have carried off the women's doubles Grand Slam in a single year, dominating at the four majors in 1984.
The other teams to have won all four are Gigi Fernandez and Natasha Zvereva, and Kathy Jordan and Anne Smith.
