The Ulster winger Andrew Trimble, who was taken off with a tight hamstring in the second half of Saturday's 32-15 victory over South Africa, has failed to recover and is replaced by Denis Hickie.
Hickie, who was drafted into the squad on Sunday as cover, has been handed the No 11 jersey for his 52nd cap, while Geordan Murphy replaces Girvan Dempsey at full-back.
Isaac Boss makes his full Test debut at scrum-half, with Peter Stringer dropping to the bench and Boss' Ulster team-mate Bryan Young also celebrates his first international start, at the expense of Marcus Horan at loose-head.
The former Wales Under-21 international Luke Tait starts in the second row for Canada against Wales in tomorrow's Test at the Millennium Stadium in Cardiff. The two nations meet in next year's World Cup.
Tait, who was brought up in Caerphilly but born in Ontario, was a member of the Overmach Parma side which knocked Newport Gwent Dragons out of the Heineken Cup in June. He is one of 11 professionals in the Canada starting side, which is captained by former Saracens scrum-half Morgan Williams.
The Cardiff Blues scrum-half Ed Fairhurst starts at full-back for Canada, with Edinburgh's Mike Pyke on one wing and Bedford's James Pritchard on the other. Fly-half Derek Daypuck, who makes his living in property maintenance, returns to the starting XV in place of Edinburgh's Ander Monro.
