Harrison, whose last-gasp line-out steal in the decisive Sydney Test guaranteed Australia a 2-1 series victory, makes way for the experienced Queensland forward Matt Cockbain.
The winger Chris Latham has also been dropped as Australia hope to bounce back from defeats to France and England. Latham's place goes to Ben Tune, who came on as a second-half substitute against France and scored a consolation try.
Elton Flatley is drafted into midfield as a replacement for Nathan Grey, who has not recovered from a neck injury.
Welsh hopes of a shock win have been hit by injury problems, with Anthony Sullivan, Dafydd James, Allan Bateman and Iestyn Thomas all out.
