The 25-year-old joins from Fulham in a deal reportedly worth £15m.
Tottenham have acted swiftly to replace Luka Modric, who was sold to Real Madrid on Monday for approximately £30m.
Dembele spent two seasons at Craven Cottage, joining from AZ Alkmaar for £5m. While injury hindered the midfielder in his first season, last term the Belgian international put in many impressive displays, attracting interest from Real Madrid and Manchester United in the process.
Dembele said on Twitter today: "Happy to confirm I signed a new contract with Tottenham FC!"
"I would like to thank all the Fulham fans for their support! You were amazing."
The arrival of Dembele does not look to be the end of Tottenham's summer transfer business. His arrival may result in the departure of Rafael van der Vaart to Hamburg, while Brazilian winger Willian of Shakhtar Donetsk and goalkeeper Hugo Lloris of Lyons are expected to arrive if deals can be reached.
Fulham are likely to push through a transfer for Sunderland's Frazier Campbell in response to Dembele's exit.
