Di Matteo underwent X-rays after limping off after a tackle from St Gallen's Daniel Imhof.
The Swiss-born Italian international has had an operation in Switzerland and will stay there for at least three days.
Assistant manager Gwyn Williams said: "Given what's happened he's as well as can be expected."
Chelsea lost the match 2-0 and crashed out of the competition 2-1 on aggregate.
