The Everton centre-back's fractured toe will be monitored by England's medical staff during the next 24 hours.
But Jagielka will have to train tomorrow at Wembley to be considered for the clash with the World Cup holders.
England are liaising with Everton's medical staff over the condition of Jagielka and spoke again this morning.
He suffered the setback against Fulham on October 23 but has continued to play by taking pain-killing injections.
The other 24 players chosen by head coach Fabio Capello were put through their paces at Arsenal's London Colney training headquarters.
It is one of the least experienced squads assembled by the Italian with 11 players having won six caps or less.
Capello has still to formally announce who will be captain after his decision not to select John Terry against Spain although Frank Lampard remains the likely candidate.
There was a poppy emblazoned on the back on the England team coach and several players wore the emblem on their boots during today's session.
Their support of Remembrance Day will be more formal tomorrow during a two minutes' silence before training at Wembley and then the players will wear poppies on their black armbands against Spain.
FIFA have agreed that the England, Scotland and Wales teams can wear poppies on black armbands during the upcoming internationals. They had turned down a request from the FA, who wanted the poppy embroidered on the England shirts.
The move came after Prince William and Prime Minister David Cameron wrote to FIFA.
