The right-back only made his debut for the club at the weekend - the 0-0 draw with West Ham.
This represents the first start for the 22-year-old who joined the club from Penarol in June 2013.
As expected, there is no Wayne Rooney which means Anthony Martial leads the attack. Behind him are Jess Lingard, Memphis Depay and Juan Mata.
Bastian Schweinsteiger and Marouane Fellaini complete the midfield.
Matteo Darmian is expected to play at left-back with Chris Smalling and Daley Blind as the central defenders in front of David De Gea.
On the bench are few options for Van Gaal, with Ashley Young the only forward with experience.
Should PSV Eindhoven win the other Group B match, which is against CSKA, then Manchester United must win tonight's game in Germany.
A loss for United will only be enough if CSKA win that other game. Should United draw, they would be out if PSV win.
