The attacker has been apprehended after ramming his car into pedestrians on a street next to a police station, injuring five including four policewomen, according to Israeli police.
Reuters reports the motorist got out of the vehicle and attempted to stab passersby before he was shot and seriously wounded by a security guard.
Police were still trying to identify the attacker. Israeli police spokeswoman Luba Samri Samri said initial reports suggested he is a Palestinian from east Jerusalem and the car he was driving was registered as belonging to a Palestinian from east Jerusalem.
Police spokesman Micky Rosenfeld said the incident occurred outside a border police station on the line separating the east and west of the city.
An investigation into the attack is underway.
Mr Rosenfeld said on Twitter: "Four policewoman injured moderately taken to hospitals in Jerusalem."
He said security in the area, which was the site of similar attacks in 2011, has been heightened.
