The collision, involving a van, a car and a lorry, occurred near Livingstone at around 8:45 on Monday morning.
The M8 is Scotland's busiest motorway.
A large plume of smoke could be seen over the accident where the van reportedly burst into flames but it is believed the flames have now been extinguished after over an hour.
Pictures posted on social media shows a white van engulfed in flames.
One person is feared dead, according to the Scottish Daily Record and two people are believed to have been taken to hospital.
The road remains closed between junctions two and three and there a long tail backs at both ends.
Traffic Scotland said on Twitter that Police Scotland was working to get all road users stuck on the motorway by the closure onto alternative routes and that they do not know when they will reopen.
A spokeswoman for the Scottish Fire and Rescue Service told the Scotsman that the first fire crews on the scene had found "numerous people in trapped in vehicles".
