Emergency services were called shortly before 2am on Thursday after the man's body was seen lying on the A40 Benhall roundabout.
Gloucestershire Police are appealing for information from anyone who was travelling on the road at that time.
Officers are making enquiries to try and identify the victim and locate anyone involved in the incident.
There is nothing to suggest that it has any connection to GCHQ, according to a police spokesperson.
A 62 year-old man has been arrested on suspicion of causing death by dangerous driving.
Motorists have been urged to find alternative routes and avoid the area due to road closures while the investigation continues.
"Diversions remain in place in Cheltenham as police investigate what appears to be a fatal hit and run collision," said a spokesperson for Gloucestershire Police.
Chief Inspector of the Tri-Force roads policing team, Alistair Barby, said: “We understand this caused a lot of disruption to road users this morning and while we are working to reopen the road at the earliest opportunity I hope people understand this is painstaking work and cannot be rushed.
“We would urge people to consider their travel arrangements ahead of rush hour and alternative routes they can take.
“In the meantime, can I appeal to anyone who was travelling on the A40 around the time of the incident and may have seen something relevant to call straight away. Likewise if you think you saw or know the victim we want to hear from you, or if you have any information about what happened. Crimestoppers can be called anonymously if you prefer on 0800 555 111.”
South West Ambulance Service said an ambulance and a rapid response vehicle were sent to the scene after being called at 1.56am.
Anyone who saw something relevant or anyone with information is asked to call 101 and quote incident 40 of 16 August.
