At around 2.30am on Friday, a car crashed into Lyons Den Fitness in the town of Bagillt in Flintshire.
The two who died were travelling in a white BMW 330 saloon when the crash took place.
Officers are appealing for anyone who saw or has dashcam footage of the BMW travelling along the A548 from the Flint direction towards Bagillt to get in touch.
A North Wales Police spokeswoman said in a statement: “Sadly, two people have died, and another three are currently in hospital in a serious condition.”
She also said: “Officers attended the scene of a single vehicle Road Traffic Collision in the early hours of this morning. The collision occurred near the junction of the A548 with the A5026 at Bagillt, and road closures were put in place.”
Two rapid response vehicles and five ambulance crews attended the scene, according to a spokeswoman for the Welsh Ambulance Service.
Police have closed a section of the A548 between Bagillt and Greenfield until lunchtime while officers continue their investigations.
Lyons Den Fitness issued a statement on social media to say that it was closed until further notice as the crash had caused structural damage to the building.
On the gym’s Facebook page, it says: “Gym closed. Sad news I’m afraid.
“Major traffic incident with structural damage to building. Honestly can’t say anymore than this ATM (at the moment).
“With the police, will keep you all updated as I know more re opening back up.”
