The actor, who has been nominated eight times in total, beat Clive Owen, Liev Schreiber, Dominic West and James Spader to win the award for Best Actor in a TV Drama.
The 55-year-old accepted his speech briefly in character, telling the audience in true Machiavellian style: “This is just the beginning of my revenge”.
He also thanked Netflix, House of Cards lead writer Beau Willimon, and his co-stars Kate Mara and Robin Wright.
“It’s a joy to come to work with your every day and all of those that I work with every day,” he said.
He went on to pay tribute to the late filmmaker Stanley Kramer in an anecdote, who once thanked Spacey for his kind words about his work but added: “I just wish my films could have been better”.
“And so I stand here tonight as someone who has enjoyed such an extraordinary career, in large measure because of the people in this room, I just want it to be better. I just want to be better, but this is very encouraging. Thank you very much,” Spacey said in concluding his speech.
Elsewhere in the TV categories, Ruth Wilson won Best Actress in a TV drama for The Affair, while Fargo picked up the award for Best TV Movie or Miniseries.
Amazon won its first award for transgender drama Transparent, which also saw actor Jeffrey Tambor win for Best Actor in a Comedy TV Series.
