It will be the Manchester rock band's only 2013 UK festival appearance.
The gig comes after the band reunited for sell-out shows at Manchester's Heaton Park last summer before embarking on a world tour attended by one million fans across Europe and Asia.
John Giddings, the festival's promoter, said: "When you talk about the soundtrack of our lives, the Stone Roses are embedded in it. I am so proud to get them to our island- they will kick the weekend off with a bang."
The four-piece, Ian Brown, John Squire, Mani (aka Gary Mounfield) and Reni (aka Alan Wren), have never played at the Isle of Wight festival before.
The festival runs from 14 June 2013. Tickets go on sale on Wednesday.
