The rapper announced he will play the venue on 11 July after finding out he was unable to play Hyde Park this summer.
It will be the first time he has played a solo London show since 2001, having previously only headlined slots at festivals in the UK.
“I heard that I wasn’t going to be approved to play in Hyde Park and it made me even more excited to play in London this summer,” he said.
“The easiest way to get me to do something is to tell me I can’t do it. Thanks to the Royal Parks for making it possible…for Wembley,” he told Zane Lowe’s Radio 1 show last night.
Eminem’s new single “Headlights” will be released to coincide with the Wembley show, with Spike Lee confirmed to direct the video.
The rapper’s latest album The Marshall Mathers LP 2 went straight to number one in the UK, becoming his seventh consecutive number one album.
His most recent single, “The Monster” featuring Rihanna, also went straight to the top of the UK singles chart after its release at the end of 2013.
Tickets for the Wembley show will go on sale at 9am on Friday 9 May.
