The Monty Python alum had been working on this film for nearly two decades due to financial issues, casting problems and a variety of delays, but the trailer finally debuted on Thursday.
The film is Gilliam's "cursed" perspective on the iconic Miguel de Cervantes novel Don Quixote, starring Adam Driver as Toby - a marketing executive who meets a man (Jonathan Pryce) who he thinks is La Mancha's defender of chivalry, Don Quixote.
The plot sparks confusion for its characters as Toby is mistaken for Quixote's squire, Sancho Panza, and the duo travel in-between the 21st and 17th centuries on an adventure.
The trailer features battles with humans and windmills, ostentatious costumes and meta references.
Gilliam's years of comedy are especially apparent when Quixote chides Toby for wanting to read his book.
"A peasant like you, cannot read," Quixote says. "I'll sound the words and you can look at the pictures."
The Man Who Killed Don Quixote will also star Stellan Skarsgård and Olga Kurylenko in main roles.
In the 1990s, Gilliam first attempted to make the film and cast Johnny Depp as Toby and Jean Rochefort as Quixote.
While there's no exact release date, it is expected to premiere at Cannes in 2018, however a legal battle might hold up its debut.
