The two famously shared the screen in the climactic Russian Roulette scene in Michael Cimino's Oscar-winning drama The Deer Hunter.
They've swapped the Vietnam War for a very different one - specifically The War With Grandpa, which Deadline reports will be an adaptation of the Robert Kimmel Smith book that follows a series of pranks between a young boy and his grandfather when the former has to give up his bedroom.
De Niro and Walken will play best friends - as they did in The Deer Hunter - and American Pie stalwart Eugene Levy will also star.
The Deer Hunter also starred John Cazale and Meryl Streep. Walken won the Academy Award for Best Supporting Actor film for his role in the film which went on to also win Best Picture.
Earlier this year, De Niro starred alongside Zac Efron in Dirty Grandpa which was maligned by critics upon its release. Last month, he took part in a 20th anniversary Q&A about Michael Mann's crime film Heat alongside his co-star Al Pacino; it was moderated by filmmaker Christopher Nolan.
